1) topology will be multi pass protein where N terminus toward outside

2) after processing of N terminus signal peptide protein sequence get exposed which is starting with methione and could binding site for certain signal molecule

3) cetainly these proteins are of eukaryotes as addition of signal peptide and it's process required endoplasmic and Golgi complex which are present in eukaryotes.
