According to the Chargaff's rules, the DNA from any cell of all organisms should have a 1:1 ratio (base pair rule) of purine bases [for the DNA cytosine, thymine and for the RNA uracil]. The amount of guanine should be equated to cytosine and the amount of adenine should be equaled to thymine.The purine always pairs with pyrimidines. Cytosine pairs with guanine and adenine pairs with thymine.

5 % adenine + 30 % guanine + 40 % cytosine + 25% uracil = 100 % DNA bases

C ≡ G 30%

T ≡ A 5%

A ≡ T 25% (Uracil is converted to Thymine in DNA)

G ≡ C 40%

100% - (5 % + 25 %) = 70% DNA base - ( Adenine % + Thymine %)

70% / 2 = 35%

35% of Cytosine .
