Staphylococcus aureus can be determined using mannitol salt agar test. S. aureus is capable of fermenting mannitol. Fermentation of mannitol is indicated by acid production. The media which contain phenol red as indicator will turns to yellow on acid production. Coagulase positive staphylococcus that is S. aureus will produce yellow colony with yellow baground indicating acid production and other staphylococcus sp. That is coagulase negative produce red colonies in red baground. Hence S.aureus can be easily identified.
