In humans, Xeroderma Pigmentosis (XP) is characterized by hypersensitive to UV light. This light creates a very high amount of DNA damage leading to increased cancer rates. You are very interested in identifying a gene that might be resulting in this UV induced DNA damage in people suffering from XP. Propose a research design that will identify, isolate, and characterize a gene involved in this disease. Include the methodology used and outcome/result you expect at each step along this research design.
First, you have to radiated the cells by UV light.
It will affect DNA damage.
Then after that you have to measure the tumour suppressor gene as well as protooncogene expression by Western blot analysis.
And previous to this state, you can see the genetic chaining pattern by restriction mapping or many other mapping which will be resulted in different band as the result of mutation and then you will see which gene expression becomes lessen by RT PCR,
And western blot and you can characterize the protein as protooncogene or tumour suppressor gene if their expression becomes increases or decreases respectively.
