In an enzyme-substrate reaction, the rate of product formation is directly proportional to the availability of active sites present on the enzyme. The more the availability of active sites, the more the product will be formed. Hence, initially with an increase in time, more and more substrates bind to the enzyme active site and get converted to product. But slowly, most of the active sites are bound to the substrate and the remaining free substrates find it difficult to bind to an available active site. Therefore, the rate of product formation decreases at the end of the experiment. We, therefore, get a bell-shaped curve in our graph.
