Ans- Steps of eukaryotic transcription involving RNA polymerase II :
1.TFIID binds to TATA box of DNA
2.TFIIA, TFIIB, TFIIF and RNA pol II bind
3.TFIIE and TFIIH bind
4.Synthesis of the pre-mRNA begins at the +1 nucleotide
5.A 5' cap is added to pre-mRNA
6.Spliceosome complexes carry out intron splicing
Transcription is the process of synthesis of RNA from DNA.
Eukaryotic transcription is occurs in the nucleus of the cell and contains three sequential stages are initiation, elongation and termination. Eukaryotes use transcription factors (like TFIID, TFIIA, TFIIB etc.) to first bind to promoter region and then help recruit the appropriate polymerase enzyme.
TFIID is the first transcription factor binds to DNA during the pre-initiation transcription complex of RNA polymerase II. TFIID binds to TATA box in the promoter region of gene, helps in recruitment of other transcription factors required for transcription.
TFIIB and TFIIA transcription factors stabilizes the association of TATA binding proteins (TBP) with the TATA box.
The presence of TFIIB allows the recruitment of a TFIIF- RNA polymerase II complex and then of TFIIE and TFIIH. TFIIE regulates the activities of TFIIH.
During intron splicing, non-coding region (introns) are removed and coding regions (exons) are joined together. Introns splicing is post transcriptional event.
