Answer #1

Answer 8)

There are found following elements in the vector:

* GFP: It stands for green fluorescent protein. This is used to detect the position of plasmid in the cell. Because there is found green fluoresecence because of this gene expression.

* Amp: These are the genes which provides the ampicillin resistance to the bacteria. This allow bacteria to grow even in presence of ampicillin.

*G418: It provides the resistance to bacteria against genticin. When this gene is not found then there is not found elongation step in protein synthesis.

*Ddp2: It stands for DNA distortion protein. 2. It maintains the structure of the DNA.

Other gene essential for plasmid:

*ori: It stands for origin of replication. This allow the replication process in the bacteria.

* Restriction sites: This are cleaved by endonuclease and allow the insertion of gene of interest in the plasmid.

A biology 207 student sub cloned the promoterless, GFP gene from pCamR-GFP into pUC18-207 using a double digest. They used competent, prototrophic DH5aE. coli cells in their transformation protocol and plated the transformation reaction onto minimal medium that contained additional supplements to help them screen and select for their desired transformants, which are transformed E. coli cells that express the GFP gene. All experimental procedures were followed such as incubation times etc. Note that GFP encodes for a protein that...
