Secretory proteins were targeted to the Endoplasmic retiuculum by the signal sequence at amino terminus,was removed duringand incorporated to the growing polypeptide chain in the ER. This wasshowed by experiments that translation of secretory protein mRNAs on free ribosomes yielded proteins that retained their signal sequences and larger than normal secreted proteins. , when microsomes added to the system, the growing polypeptide chain are incorporated into the microsomes and the signal sequences are removed by the proteolytic cleavage.
