Avery, Mc Cleoid, and Mc Carthy provided biochemical evidence to state that nucleic acids are the genetic material. They have used different hydrolytic enzymes (Glycosidases, proteases, lipases, and nucleases) to prevent transformation. Transformation is inhibited only in the presence of nucleic acids.
R strain = Non-pathogenic due to the absence of an outer glycocalyx
S strain = Pathogenic due to the presence of an outer glycocalyx

S-strain can make one or more enzymes that can synthesize the glycocalyx which helps in immune evasion
