The region of FK1 needed for nuclear export has a motif LRxSxSTxTxP with several serine and threonine residues (x = any amino acid).

Test for weather the protein has some site for phosphorylation ot not can be tested by site directed phosphorylation in each of these specific amino acid residue then followed by treatment with kinase enzyme which can abke to incorporate phosphate group the phosphate must be tagged with Radiolabbled which can be identified by autoradiography.

If the amino acid sequence is a target form phosphorylation then the Radiolabbled phosphate cannot be identified as as other amino acid residue are blocked with phosphorylation. So,Radiolabbled sigbal will be observed. Thus, phosphorylation inhibition assay can be used for detection of site specific phosphorylation withen a sequence of a protein
