Ans.4) Expected result of LB only plate – After adequate incubation, the agar medium demonstrate the development as it is confirmed by colony formation. In the broth medium, development is apparent by the presence of turbidity. The cells grown on LB media do not transformed as they do not have ampicillin resistance gene.

5.) The plausible reason for no growth occurrence on LB only plate is that it did not contain the ampicillin resistance gene. This may occur due to bacteria was killed by the ampicillin.

6.) Positive controls displayed transformation with presence of ampicillin resistance gene as maker. The transformation is successfully carried out and it is confirmed by expression of GFP gene. Hence, tee last plate LB+AMP+ARB is considered as positive control.
