Casein is a milk protein. Casein gives an opaque color to the milk agar plate.
If the bacteria is metabolized casein, it secretes casease which degrade casein protein into smaller peptides.
When the bacterium, which was grown in the milk agar plate metabolize casein, turn the opaque color of milk agar into a clear plate. change in color from opaque to colorless shows the bacteria is metabolizing casein.
