Yes, it is possible.

Sender :

A sender with a 2-tier architecture uses the user-agent( a software agent) to interface with his/her email server, which will further send out web-based SMTP packets to the receiver’s end server computer containing a mail server software.

Reciever :

The server at the receiver's end will issue an IMAP or SMTP packet to the receiver’s server computer, which will be accessed by the receiver when he/she requests for the email with an HTTP request to the web based email application.

Thus, a 2-tier architechture easily interfaces with a 3-tier architecture using the appropriate protocols.
