Which of the following pentapeptides will exhibit the highest extinction coefficient at 280 nm? Assume that each tryptophan has an extinction coefficient of 5690 M-1cm-1, tyrosine has an extinction coefficient of 1280 M-1cm-1, and the absorbance of phenylalanine at 280 nm is negligible.

Answer #1

W is tryptophan and Y is tyrosine.

Tryptophan and tyrosine has extinction coefficient of 5690 and 1280 M-1cm-1.

The pentapeptide with larger number of this two will have greater extinction coefficient.

TYWSF has 1 tryptophan and 1 tyrosine.while RYYFE has two tyrosine and FEWFE has 1 tryptophan.

Thus,TYWSF has highest extinction coefficient due to both tryptophan and tyrosine.

RYYFE has two tyrosine, so the extinction coefficient is 1290×2=2580 M-1cm-1.

We know, A=€cl

0.05=2580×c×1.

c=0.05/2580

c=1.94 E-5 M

c=19.4 micromolar.

    Human myoglobin is a protein with a molar extinction coefficient at 280nm of 1.40 x 104 M-1cm-1 . 100.0 uL of the stock solution of myoglobin (the tube was marked as 0.050 mg/mL) was mixed with 0.900 mL of buffer, giving an absorbance at 280nm of 0.810 (cuvette is 1.00 cm pathlength). From this information, what is the molecular weight of human myoglobin? Do you think this is a reasonable value? Explain WHY or WHY NOT?

    Please answer both 1. Often when handling proteins, we use concentration in terms of mg/ml. A high concentration is greater than 1 mg/ml and low concentrations are below 0.1 mg/ml (roughly). If you obtained a sample of MDH that was 10 micromoles/L, what is the concentration in mg/ml? Assume that the molecular weight for MDH is 32 kDa. 2.
