Answer #1

BAPNA stand for-alpha-benzoyl-L-arginine-p-nitroanilide.This is a synthetic substrate contains a dye which is bound with an amino acid.

BAPNA test use to detects the enzyme activity usually activity of teypsin in the sample. Teypsin is an endopeptidase , its cleave the peptide bond between amino acid and dye .ctgus release the dye and solution colour turns to yellow .this is an indicator of enzyme activity.

1. BAPNA+ TRYPSIN - solution colour turns to yellow

2. BAPNA+ PMSF+ TRYPSIN - solution colour does not change

This PMSF (phenylmethane sulphonyl flouride ) is an

Protease inhibitor, this inhibits activity of trypsin thus due will not released from BAPNA.

3. BAPNA+ STI+ Trypsin - colorless solution

As STI stand for soybean trypsin inhibitor, inhibits trypsin thus

Dye will not release colour of solution will not change.
