Both Gal4/UAS and Repressor/operator involve the relief of expression in the presence of the induce (Galactose or lactose respectively). What is the kay difference between the two systems?

The basic difference between the two given operons lies in terms of the way in which they bring repression. In Gal operon, the repressor molecules bind to two operator sites. This is followed by the dimerization of these repressor molecules which leads to the creation of a DNA loop that decreases the accessibility of DNA to RNA polymerase. However, in case of lac operon, a single repressor binds to the operator and does not allow RNA polymerase to bind. Thus, there is no formation of DNA loop in lac operon.
