Proton pumps play key role in cellular respiration.Free energy released by oxidation of glucose to carbon dioxide is is retained in reduced coenzymes NADH and FADH2 , generated during glycolysis and Kreb's cycle.Electrons from NADH and FADH2 are transferred to O2via electron transport chain,The electron transport chain includes three complexes( I, III, IV ) that are proton pumps.As high energy electron is passed through the electron transport chain, some of the energyreleased is used by three complexes ( I, III, IV ) to pump proton out of the matrix space.The resulting electrochemical proton gradient drives back proton into mitochondrial matrix through ATPase thereby generating ATP. Blocking proton pumps would inhibit electron flow, and hence generation of ATP.The consequence would be inhibiting complete oxidation of glucose to ATP and CO2 .Failing to meet energy demands of the cell and inhibition of cellular respiration would be lethal to the organism.
