When the regulatory region of the VND6 gene is fused to a GFP marker gene and a terminator and the construct introduced into plants, GFP is expressed in immature vascular cells. When the transcription unit of the VND6 gene is fused to a regulatory region that provides expression in non-vascular mesophyll cells in transgenic plants (i.e. misexpression), these cells turn into vessel elements that are normally found only in vascular tissues and are used for water transport. Based on these experiments, what is the function of the VND6, a. regulatory region? b. transcription unit? c. gene?

You are studying the regulatory DNA of a mouse gene expressed in developing heart, liver, and lung tissue. Your preliminary work has shown that heart and lung expression of this gene is controlled by a short fragment of DNA just upstream of the promoter. Based on this result, you decide to investigate this region further to understand its function. You decide to compare this sequence to the regulatory DNA of the same gene found in rats and humans. Using genome...

You are conducting an experiment identifying enhancers that regulate the expression of a gene that codifies for a protein that participates in cell division. The gene is called Mitosis Regulatory Protein A or MRPA. The complete DNA sequences for the MRPA promoter and coding region have been identified. However, it is unknown if MRPA has enhancers regulating its transcription. To answer this question your lab first produced a transgenic cell line where GFP has been inserted as a reporter gene....

molecular biology Section C (40 marks) Answer ALL questions from this Section 5. You have isolated total RNA from muscle cells and constructeda muscle cDNA library. You wish to study the regulatory region of a muscle-specific cDNA gene (gene M) that you have previously identified. 6 (a) For your study, you need to isolate a genomic clone of gene M. Why isa cDNA clone of gene M not appropriate for your study? (2 marks) (b) Outline the steps you would...

What control elements regulate expression of the mPGES-1 gene? The promoter of a gene includes the DNA immediately upstream of the transcription start site, but expression of the gene can also be affected by control elements. These can be thousands of base pairs upstream of the promoter, grouped in an enhancer. Because the distance and spacing of these control elements make them difficult to identify, scientists begin by deleting sections of DNA that contain possible control elements and measuring the...

Hagrid has purified a protein from unicom sweat and called UNI-1. He has then doned the Unit gone and inserted in rat zygotes (by microinjection) a recombinant construct containing the Uni-1 coding sequence its entire 5 UTR region, covering both the minimal promoter and a series of cutative regulatory als elements. The mutantrats develop glorious, luminous hair. Hagrid has also discovered that the UNI-1 protein binds very tightly to DNA and hypothesizes it is a transcription factor that regulates proliferation...

    2. A dominant allele H reduces the number of body bristles that Drosophila flies have, giving rise to a “hairless” phenotype. In the homozygous condition, H is lethal. An independently assorting dominant allele S has no effect on bristle number except in the presence of H, in which case a single dose of S suppresses the hairless phenotype, thus restoring the "hairy" phenotype. However, S also is lethal in the homozygous (S/S) condition. What ratio of hairy to hairless flies...
