Lane 1 has Eco R1 and Sa2 digestion. There is only two restriction site that's why two bands are produced.  

Lane 2 has hind3 and sa2 digestion. There is also two bands produced.

Lane 3 has ECoR5 , Sa2 and Eco R1 digestion. There are three restriction sites. Hence 3 bands produced.
