The immune system is self-restricted and the self-restriction is seen with immunological responses that involve both the MHC class I and the MHC class II molecules. Define MHC class I and class II restriction, which molecules and cells are involved and which stage of the immune response (stages 1 – 8 in figure 1-7 on page 20) the class I and class II restriction occurs (pick a stage from figure 1-7, state that stage, and describe the MHC restriction taking place (class I or II and the cells and molecules). Then describe an experiment which shows MHC class I restriction and then an experiment showing MHC class II restriction. Be sure to define, describe and name all antigens, animals, cell, purification systems and assays (readouts) used.

  • Definition of MHC class I and class II restriction and the molecules and cells are involved in these classes:

MHC restriction where the APC or target cells express MHC molecules that the T cell recognizes as self in order for T cell to respond to the antigen presented by that APC or target cell.

CD8 T cells bind class I MHC which are expressed on most cells in the body.

CD4 T cells bind class II MHC which are only expressed on specialized APCs.

  • The stage of the immune response where the class I and class II restriction occurs: In the effector phase. In this phase of the cell-mediated response, activated macrophages carry out phagocytosis and cytolysis. Interruption of any of these steps results in significant cellular immune suppression.
