1) That restriction site is a SNP.

That restriction site is a RFLP.

Explanation: Sometimes, SNP in a genome can make a site as a restriction site, which was not, before the SNP.

2) Three gel bands would the probe above detect on a Southern blot of an individual heterozygous for the polymorphic restriction site.

Explanation: As the individual is heterozygous, he would have one gel band that does not contain the same pattern of restriction site. So, the probe can hybridize with that. The second DNA that has the above mentioned restriction site pattern, would generate more than four fragments after restriction digestion. Among them two fragments will hybridize with probe.
