Transcription by RNA Pol II consists of several stages.They are initiation, elongation, promoter clearance, elongation and termination. pTEFB is a cyclin dependent kinase, that regulates the elongation phase of transcription.It controls elongation by phosphorylating  RNA Pol II and antagonizing the effects of negative elongation factors.Elongation step is critical for generating full length M RNA transcripts and also plays a part in regulation of pre M RNA capping, splicing and polyadenylation.. So if a cell line loses pTEFB activity, elongation of M RNA cannot take place and as result M RNA transcripts will not be available for gene expression.
