You want to track translation of your transcription factor within a pancreatic cell for your Excellence in Research project. You perform a pulse-chase experiment, where radioactive amino acids are introduced to the cell over a short period of time (pulse) and then are washed away (chase). The radioactive amino acids will be introduced into newly synthesized protein and you can track the radioactive proteins as they are sorted throughout the cell. You also add a drug that prevents translation of all proteins except your transcription factor. Where would the first radioactive signals appear after you wash away the free radioactive amino acids?

1. The Golgi

2. The cytoplasm

3. Both the ER and the cytoplasm

4. The nucleus

5. The ER

Answer #1

radioactive amino acids are given, also a drug that inhibits translation of proteins other than transcription factor are also given so only transcription factors will incorporate radioactive amino acids transcription factors localize to the nucleus to carry out the transcription of genes so when free radioactive amino acids are washed away signal will come from nucleus only, so the answer is 4) the nucleus

Scientists can track the movement of proteins through the endomembrane system using an approach known as a pulse-chase experiment. This experiment involves the “pulse” phase: Cells are exposed to a high concentration of a radioactively labeled amino acids for a short period to “tag” proteins that are being synthesized. the “chase” phase: Any unincorporated (not used to make proteins) radioactively labeled amino acids are washed away and large amounts of the same, but unlabeled, amino acid are added. Only those...

When a protein is co-translationally translocated into the ER it sometimes receives attachments of eight mannose residues, which are added to a core carbohydrate unit consisting of GlcNAc (N-acetylglucosamine). The mannose residues are trimmed down from a unit of eight to a unit of five either in the ER prior to transport to the Golgi, or in the cis-Golgi after transport The removal of three mannose residues can be detected using gel electrophoresis because proteins modified in this way will...
