ANSWER

Phenol-Sulfuric Acid Method is most reliable method for carbohydrate estimation. it is mostly use in measuring neutral sugar content in oligosaccharides, proteoglycanes, glycoproteins and glycolipids.

In hot acidic medium, glucose is dehydrated to hydroxymethyl furfural. This form yellow-brown coloured product with phenol and its absorption maximum at 290nm. t204 HO -3 40 4

Here 5% phenol is being used as regent and for the method while 90-95% sulfuric acid is used.

Yes a standard curve needed, The phenol sulfuric acid methode is carried out by preparing a set of solution with known glucose concentration and mixing them with the phenol- sulfuric acid regent. A standard curve can be made and the concentration of unknown sugar sample can be derived from the standard curve.
