Reticulocytes appear blue-gray color on the wright stained smear.In this stain, only the early reticulocytes with the most residual RNA will be stained (that is only those cell turns bluish in color called as polychromatophil).

Methylene blue stain is designed to highlight the RNA as blue color against an otherwise pink cell. Depending on the stage of the reticulocyte, the blue color is distributed in general(early reticulocyte) or punctuate (late reticulocyte) pattern.

In both cases reticulocyte containing the residual RNA is being stained but the difference is unlike in wright's smear, Methylene blue stains both early and late reticulocytes.
