1. During the test antigen- negative blood is given to the patient for safe transfusion. In negative reaction , the pellet passes easily to tge bottom, as no agglutination occurs and is scored as "0".

Agglutination is graded on a scale from 0 to 4+.

4+ reaction = RBCAs ( red blood cell agglutinates) remain at top of gel.

3+ reaction= RBCAs in top half column

2+ reaction = RBCAs scattered through out the column

1+ reaction= RBCAs at lower half of the column

0= no agglutination and red blood cell pass all the way to the bottom.

So here , as per the screening test the patient has O+ve blood group since , anti A and anti B agglutination is graded "0".

2. Auto antibody is an antibody produced by immune system that direct towards individual own proteins egs : ANA , anti - ds DNA

Alloantibodies are immune antibodies produced only after exposure to foreign RBC antigens.they react with allogenic cells when exposed during pregnancy and transfusion..

So, here specificity of this antibody is alloantibody.

3 . In immediate spin the reporting is 2+ and at 37 degree celsion 1+ reporting is present,so the screen is reported as positive and an antibody panel should be performed to determine the specificity of the antibody present.
