Answer #1

A#5: When all of the sample to collected has left the column the drop of solution coming of the tip is neutral because until the all of the sample mobilizes through the ion exchange column all the Cu2+ ions would bind to the resin and release H+. When the sample left the column (no sample) there will not be any exchange as a consequence H+ will not be liberated any more. Thus the solution coming of the tip would be neutral (PH = 7 the value of deionized water).

A#6: Cu2+ has 2+ charge and H+ has +1 charge. Therefore, a single Cu2+ ion will take the place of 2 H+ ions. The answer is 2 H+ ion will be dispalced by 1 Cu2+ ion in the ion exchange column.

A#7: The ion sites are negativey charged and they are saturated with H+ ions before the sample run.

A#8:Ions are displaced from the resin binding sites by any ion that has higher binding affinity.

SIRJE GEL OBJECTIVE: Partial purification of a protein, isocitrate dehydrogenase (IDH), using DEAE ion exchange chromatography. IDH will be eluted from the column using increasing concentrations of KCI. IDH will be tracked using the IDH activity assay and the extent of purification will be analyzed using SDS PAGE Last week we assayed for the activity of the enzyme, isocitrate dehydrogenase, from E.coli strains carrying a plasmid that over-expresses the E. coli IDH gene. To purify proteins from cel extracts, biochemists...

In this lab exercise a solution containing K+ is passed down an ion exchange column containing a cation exchange resin in the acid form. The K+ ions in the solution replace the resin bound H3O+ ions which elute from the column and then are titrated. This means that the column now holds the K+ ions. To use the column again we must "recharge" the column to remove K+ ions. In your Lab Notebook, describe the exact procedure you will follow...

Acetylation of ferrocene and column chromatography Lab 4: Question 3 Homework • Unanswered Important ... The solvent is changed from petroleum ether to diethyl ether after the ferrocene is collected from the column. Why not use diethyl ether the entire time? Because diethyl ether is more polar than petroleum ether Because petroleum ether selectively elutes ferrocene since it contains more ether oxygens than diethyl ether Because diethyl ether would lead to a poorer separation of ferrocene and acetylferrocene Both a...
