Topoisomerase are enzymes that plays an important role of maintenance during DNA replication in the overwinding or underwinding of DNA. Type I topoisomerase act by changing DNA twist. This enzyme are not able to unknot or untangle duplex molecules. To unknot or untangle duplex molecules an enzyme is required another enzyme that can act on DNA writhes.
Type II topoisomerases act on DNA writhes that can remove knots and tangles from the DNA.
So topoisomerase-I can change the twist number and topoisomerase-II can change the writhe number.
