Why must mineral oil be used with our PCR machine?
Mineral oil is used when the PCR machine does not have a heated lid. Mineral oil serves to prevent the evaporation of liquid and the change in composition of salts and buffer due to evaporation and condensation which might affect the enzyme activity. In order to prevent changes in the volume of the reaction mixture in the absence of a heated lid, mineral oil is used.
How and why are positive and negative controls used for the PCR?
50 ml of mineral oil is mixed with a liter of water and is subjected to high shear to create 100 nanometer spherical oil droplets. Calculate the new surface area of droplets in square meters and the number of droplets in the resulting emulsion. Assume the density of mineral oil to be 0.85 gms/cc.
1a. Because his patient is allergic to mineral oil, a physician has asked you to substitute an equal weight of olive oil for the 10 ml of mineral oil he has in a lotion. How many milliliters of olive oil would be needed? Specific Gravity of mineral oil is 0.85 and Specific Gravity of olive oil is 0.91 Ans:_____mL 1b. Rx: Tannic Acid (20.0g)/ Exsicc. Sodium Sulfite (0.2g)/ Glycerin (25g)/ Yellow Ointment (54.8g)/ Total= 100g How many mL of glycerin...
Why are we treating our purified PCR products with a DNA polymerase before adding them to the ligation reaction? and What is the purpose of ATP in the ligation reaction? Group of answer choices -It provides energy to the ligase to catalyze a peptide bond -It provides energy to the ligase to catalyze a phosphodiester bond -It provides energy to the ligase to cut the DNA -It provides energy to the ligase to unwind the DNA
The price of oil is a very important and volatile resource in our economy. Oil has many different functions that are either used as a final or intermediate good for our economic functioning. How do you think the United States should deal with the vulnerability of importing oil? What impact does OPEC have on the price of oil when the US, one of the largest importers of oil, is experiencing either a recession or economic growth? Why would a recession...
In a hypothetical experiment, a student performs an extraction by shaking 25.0 mL mineral oil with 50.0 mL of 1.00 M I2 aqueous solution. After allowing the layers to separate the student observes that the mineral oil layer has become purple in color, indicating that some of the I2 has been transferred from the aqueous layer to the mineral oil layer. The student then removes the aqueous layer and titrates it with 5.00 mM Na2S2O3 solution. At the endpoint of...
How is PCR used in DNA cloning? Can you provide some real life examples of PCR cloning? Why/When do we use PCR over alternative cloning techniques.
A number of different buffers/components are used in the PCR. Explain the function of each: a. DNA plasmid b. Primer pair C. dNTPs mixture d. Taq polymerase Question 2 (3 points) In our PCR reaction the optimum annealing temperature is 56°C. Explain the general factors that help determine the optimum annealing temperature of a PCR reaction.
Assume that you are using an open-end manometer filled with mineral oil rather than mercury. The level of mineral oil in the arm connected to the bulb is 243 mm higher than the level in the arm connected to the atmosphere and atmospheric pressure is 741 mmHg. What is the gas pressure in the bulb in mm of Hg? (The density of mercury is 13.6 g/mL, and the density of mineral oil is 0.822 g/mL.) Express your answer with the...
