5) At position 2*, a substitution is observed because A is being replaced by T. Since A is a Purine and T is a pyrimidine base, so Purine <---> Pyrimidine substitution is called Transversion.

6) The correct answer is false.

Reason : PCR amplification and subsequently gel electrophoresis of 1* position will not help in any way to detect variations. This is because of the fact that Gel electrophoresis only works when we want to differentiate between DNA fragments of different lengths. Gel electrophoresis will give different bands, based on the size of DNA fragment being amplified. But here, the 1* position of the query sequences are of equal size. So whatever will be the composition of blank spaces, electrophoresis will not be able to detect it. It can only tell about the length. The length of DNA fragment and it's composition are two independent things. So we cannot find any variations if we do gel electrophoresis.
