Transcription initiates, when a) sigma factor recognizes and binds to upstream -10 and -35 sequence elements of the target genes.

RNA polymerase holoenzyme consists of core enzyme and sigma factor. Sigma factor is formed of a single protein chain, loosely attached to core enzyme. The sigma subunit enables RNA ploymerase to recognize promoter region of DNA. It recognizes specific transcription initiation sites, where RNA synthesis can begin. These sites are called promoter sites. Sigma-2 region recognizes and binds to -10 region (5'-TATAAT-3'), whereas sigma-4 recognizes and binds to -35 region (5'-TTGACA-3').
