Membrane filtration allows only the microbes that are of a specific size to pass through. If the diameter of the membrane filter is made so narrow that only a single microbe can pass through it, then by placing a laser each microbe can be counted by using the membrane filter while it passes through the filter. In this way there is no use of spectrophotometer.
