Answer #1

Dialysis experiments are based on the fact that the final concentration in the whole mixture of sample and dialysis buffer is the same. When the solution reaches equilibrium, the concentration of imidazole can be calculated as:

C_{eq}=\frac{C_{i}\cdot V_{i}}{C_{eq}}=\frac{0.300M\cdot 3.00mL}{503.00mL}=0.00179M

This will be the concentration in the sample once equilibrium is reached.

    A. What is the Isoelectric point pI of the peptide Gly-Glu-Ala-Lys-Val ? B. A purified protein is in a Hepes buffer at pH 7 with 300 mM NaCl. A sample (2mL) of the protein solution is placed in a tube made of dialysis membrane and dialyzed against 1 L of the same Hepes buffer with no NaCl. Small molecules and ions (such as Na+, Cl-, and Hepes) can diffuse across the dialysis membrane, but the protein cannot. 1. Once the...

If dialysis is allowed to proceed to equilibrium before each change of dialysate buffer, the substances retained by the membrane are purified by a factor equal to the ratio of buffer volume to sample volume. If 5mL of a solution contains 1mol/L NaCl is placed in a dialysis membrane and then placed in 500 mL of water and allowed to reach equilibrium, what will the final concentration of NaCl be within the dialysis membrane? If the dialysis membrane above is...

From Hung et al., (2017) “Purification, characterization and biological effect of lectin from the marine sponge Stylissa flexibilis (Lévi, 1961)” Specimens were cut into small pieces, ground with a mixer machine and extracted (1:2, w/v) with 50 mM Tris–HCl, 150 mM NaCl buffer (pH 7.5) for 6 h at 4 °C. After filtration through a cheese cloth, the filtrate was centrifuged at 3500 × g for 30 min at 4 °C. To the supernatant, cold absolute ethanol (− 20 °C) was...

It's a weak acid strong base titration Experiment 4: Identification of an unknown acid by titration Page 2 of 15 Background In this experiment, you will use both qualitative and quantitative properties to determine an unknown acid's identity and concentration. To do this analysis, you will perform a titration of your unknown acid sample-specifically a potentiometric titration where you use a pH meter and record pH values during the titration, combined with a visual titration using a color indi- cator...
