Can you please explain what the right answer and why?
43. You are using an anti-mouse monoclonal antibody to a nonlinear conformational epitope in an ELISA. Which of the following is TRUE? This antibody should work well in a western blot from an SDS-PAGE gel This antibody cant be used in a western blot because it will be denatured by the residual SDS This antibody be useful in targeting a protein blotted from a native gel. This antibody should have higher affinity than a polyclonal antibody. This antibody will target only human proteins a. b. c. d. e.

Answer #1

Answer is c. This antibody be useful in targetting a protein blotted from a native gel.

In general, the primary antibody that recognizes the target protein in a western blot is not directly detectable. Therefore, tagged secondary antibodies are used as the means of ultimately detecting the target antigen (indirect detection). Primary antibody is an unmodified mouse monoclonal antibody, then the secondary antibody must be an anti-mouse IgG secondary (or non-IgG) antibody obtained from a non-mouse host. Monoclonal antibodies (mAbs) recognizing native epitopes are an important tool for functional and structural studies of proteins.
