4- Codons are made up of three nucleotides. Some amino acids such as leucine, serine are coded by more than 1 codon which has a difference of the last nucleotide.

So if we were to have a mutation in the nucleotide in a codon then we will pick the last nucleotide as it will be least damaging.

6- mRNA is read in 5' to 3' direction. 5- end of the mRNA has the shine-Dalgarno sequence in case of prokaryotes and Kozak seuqnece in case of eukaryotes which allows ribosome to binds to the mRNA and translate it into protein
