No, the idea of reproductive isolation easy to test in the natural enviroment.
Because for natural environment lot of factor will affect the environment and also we need to wait for long time period to study that evolution is occuring or not due to reproductive isolation.

We can able to test in lab. But the validity of our conclusion is some what relate to the environmental factor. In actual environment the factor might be change compare to lab environment so the result might be vary.
