Why is it a problem to have EDTA during DNA sequencing techniques?

EDTA is a MG2+ Chelator, and Mg2+ is extremely crucial in stabilizing the Polynucleotide Chain by two interactions.

1. It is a cofactor of the DNA Polymerase Enzyme

2. It helps to stabilize the Polynucleotide Chain as is shown below. The phosphates are negatively charged and need to be neutralized during sequencing.

Hence EDTA is a problem for sequencing. In fact, a 1:1 mixture of the two inhibits all activity of the Polymerase! The reaction is : Mg2+ + EDTA4- → MgEDTA2-
