You notice that Alice wanted to check the cell number after one hour of growth, so she also used a Petroff-Hauser chamber (Hemocytometer) to count the number of cells. If she had
10^4 cells in the original culture tube after one hour, how many microliters (uL) of the original undiluted culture would she need to be able to count approximately 100 cells?

Answer #1

If the culture tube has 10^4 cells, she needs 100 cells or 10^2 cells, that means she has to take 10^4/10^2 = 100 times less volume of the original culture in which the cell number is 10^4. or she can dilute the original culture to 100 times to get 100cells.

Say 1ml (= 1000 uL) volume contains 10^4 cells, how much volume should contain 10^2 cells?

10^4 cells per 1000uL, so 10^2 cells per 10uL.

So if she takes 10uL of 1ml of the undiluted culture having 10^4 cells, she will get 100 cells.

Similarly if she takes 1uL of 100uL of the undiluted culture having 10^4 cells, she will get 100 cells.

Hello, just wanted to check my answers. Answer as many as you like/can. Thankyou. 1) Four grams of soil are added to 36 ml sterile water and mixed well. 0.1 ml of this mix was added to 9.9 ml sterile water. This was diluted by 4 successive 1/10 dilutions. 1.0 ml from the last dilution was used to prepare a pour plate. After incubation 289 colonies were counted. Calculate the # CFU/g soil. 2) A 0.1 ml aliquot of a...

please help with whatever possible. thank you so much in advance. Name One use of serial dilutions is to calculate the concentration of microorganisms. Since it would usually be challenging or even impossible to actually count the number of microorganisms in a sample, the sample is diluted and plated to get a reasonable number of colonies to count (usually between 25 to 250 colonies is the goal). Since each colony on an agar plate theoretically grew from a single microorganism,...

Prologue: Alice Kaninchenbau was a 20-year-old junior at Boise St. University, where she majored in Political Science and played as an open-side flanker on the women's rugby team. In the last game of the season (against Weber St.) she suffered a broken nose (and the team suffered a 0-36 loss). Always an optimist, Alice considered her broken nose to be the perfect reason to have a rhinoplasty (and get the nose she had always wanted). The case: The antibiotic erythromycin...

    10. Write a one-page summary of the attached paper? INTRODUCTION Many problems can develop in activated sludge operation that adversely affect effluent quality with origins in the engineering, hydraulic and microbiological components of the process. The real "heart" of the activated sludge system is the development and maintenance of a mixed microbial culture (activated sludge) that treats wastewater and which can be managed. One definition of a wastewater treatment plant operator is a "bug farmer", one who controls the aeration...
