DNA polymerase III is a main replicating enzyme because of its high polymerization (1000 nucleotides per seconds) and processivity rate. It has four components- catalytic core component, dimerization componenet, processivity component and a clamp loader. Catalytic component consists of three subunits 5'-3' polymerase activity, 3'-5' exonuclease activity for proofreading and the which enhace the proofreading activity. The 3'-5' exonuclease activity is responsible for proofreading which enhance the accuracy of the DNA synthesis.
