Why does the OD value stay high while the viable cell count drops drastically at 540 - 600min? How does this reflect the importance of using viable cell count over OD values when determining growth rate?

Answer:

As the number of cells in culture increase, the OD of the sample will also increase. If you are looking at an actively growing culture in exponential (log) growth, then the OD (turbidity) of the culture is a good estimate of the viable population count. If your cultures have entered lag phase however your total counts (OD) will be considerably higher than the viable number of cells in the culture since both dead and live cells are present.

Viable cell count gives exact number of viable cells at all conditions, while OD determines the cell density irrespective of live or dead
