Mannosidase II is a Golgi complex marker. It identifies the medial and trans-Golgi complex in cells. FLAG was attached to the parkin protein in this experiment. Parkin is ubiquitin ligase that helps to attach ubiquitin to on substrate proteins, and targeting them for proteasome mediated protein degradation.

In Figure 2A, parkin overexpression is seen to be co-localized with mannosidase II expression in COS-7 cells. The expression of parkin is studied by immunostaining with FLAG antibody while expression of mannosidase II is studies by using antibodies tagged with red fluorescent dye to this enzyme. If both proteins who localization at the same spot, a yellow red fluorescence is seen (part III of A) as shown by the arrow.

The gamma adaptin is associated with clathrin coated vesicles in trans Golgi Network. Hence, although their expression can also be correlated with Parkin expression in Golgi apparatus, it is mannosidase expression which will be more specific. Mannosidase II is a glycosyl hydrolase which is involved in N glycosylation pathway.

Figure 2 A will indicate parkin overexpression occurs in the Golgi. However, even Fig 2 B will indicate that parkin is expressed to a certain extent in Golgi Complex. However, the cell expression of parkin looks diffused. Further, there is less yellow red fluorescence.

Sub panels C and D are for ER and lysosome expression, while E is for parkin antibody specificity.

If only one subpanel is to be selected, Fig 2A will be more specific.
