Answer #1

Following procedure was undertaken to prove the above statement:

  1. Using a frog egg a "cycling" extract was prepared to initiate the early development.
  2. Using low levels of RNAse mRNA was depleted from this extract. This extract did not cycle.
  3. Hence, cyclin B mRNA was added to the depleted extract, after RNAse inactivation and cycling was restored.

You are a studying the progression of cells through the cell cycle. You are particularly interested in the role of the APC, which in a ubiquitin E3 ligase, in the control of the metaphase cyclin (cyclin B) and the timing of events. To look at this you start by synchronizing your cells in G1 and examining the levels of Cyclin B over time. The APC usually works on cyclin B during anaphase. RBX57 is a substrate for Cylcin B/CDK2. Given...

help please!! 21) "The paradigm that the adult heart is a post mitotic organ was established in the 1970s. Later, this concept was changed because of the growing body of evidence challenging this traditional view. In adults, myocardium can regenerate from the cardiac stem cells that enter the cell cycle, contributing to changes in the myocardial mass. A low level of cardiomyocyte renewal may occur throughout life, which may be in response to cardiac stem cell activity or migration of...

Hi! I am having trouble answering these questions, more specifically I am stuck on part B! Could you please help me out and let me know how to answer this? Thanks! 2) As a continuation of the experiments in Question 1 you want to study the timing of the APC's activity more distinctly. To do this you treat your cells with MG132- a proteasome inhibitor. When MG132 is added to cells proteins can still be ubiquitinated, but they are not...
