Answer #1

The yeast methyltransferase protein, repairs both types of damages

The mispairing of O6-methylguanine (O6-MeG) with T and O4-methylthymine (O4-MeT) with G gives rise to GC-to-AT and AT-to-GC transitions, respectively.

methyltransferase repairs O6-MeG by a suicide reaction that irreversibly transfers the methyl group to a cysteine residue in the enzyme active site to generate S-methylcysteine. However the protein has much lower affinity for O4-MeT than for O6-MeG in vitro, O4MeT repair in vivo can be inferred because expression of methyltransferase in E. coli prevents methylation-associated AT-to-GC transitions.

Can you please explain in detail DNA repair mechanisms from a biochemical point of view (direct reversal of damage- DNA methyltransferase, DNA photolyase, base excision repair- DNA glycosylase and nucleotide excision repair, as well as mismatch repair and SOS response and recombination repair) I really need your help. I will be forever grateful. And if I may ask you one thing if you copy from the internet please put the citations, or links. Thank you in advance!!!

Repair of DNA damage from UV radiation DNA damage from UV radiation needs to be repaired if DNA replication occurs without error. the following plot shows how cell survival is affected in the presence of UV radiation. Notice that the wild-type cells can overcome the damaging effects of UV radiation on DNA and show high survival even as UV increases. However, cells deficient in certain genes display relatively much lower survival rates. 1. the fact that the double mutant shows...

Part 1. Eukaryotes use at least two distinct mechanisms to control gene expression by altering the structure of chromatin around a particular gene. One of these mechanisms is the covalent modification of histones to switch DNA between an open and closed confirmation. A second mechanism is the covalent modification of DNA (typically on cytosines) by methylation. A. Describe a type of histone modification and explain what effect is has on chromatin, and how that effect is achieved. a. One type...
