  1. Formation of the strain of S. pneumoniae that was antibiotic resistant and resistant to the host immune response was the result of acquisition of a capsule forming gene by transformation.
    1. What membrane bound structure formed that increased bacterial binding to DNA by 1000X and aided in the entry of the DNA to the cell?
    2. During what part of the growth curve (lag, exponential, stationary, death) would the bacteria typically form this structure?
    3. What type of signal bound the sensor kinase?

Answer #1

Prokaryotes and cytoplasmic membrane .

During this period, called the lag phase, the cells are metabolically active and increase only in cell size. The lag phase is the log phase, in which population grows in a logarithmic fashion. As the population grows, the bacteria consume available nutrients and produce waste products.

membrane-bound sensor kinase and a DNA-binding response regulator. In response to specific stimuli, the sensor kinase is phosphorylated at a conserved histidine residue and then the phosphoryl group is transferred to the conserved aspartate on the response regulator. So, Two-component systems typically consist of a membrane-bound histidine kinase that senses a specific environmental stimulus

Recently an antibiotic resistant strain of bacteria was found to possess a plasmid containing a unique drug resistance gene. Scientists and physicians are extremely concerned about this discovery since this drug resistance gene is usually only found as a part of the bacterial chromosome (nucleoid). In 1 bullet point --Explain the differences between chromosomal DNA (nucleoid) and plasmids. In 2 bullet points -- Explain why the medical community is so concerned about the the discovery of this particular antibiotic resistant...

Frederica wants to try using yogurt as the delivery mechanism for her vaccine. She will clone a gene from S. pneumoniae and express that gene in Lactococcus lactis, an organisms used to make yogurt. Eating the yogurt would serve as the vaccine delivery mechanism. BACKGROUND: Mycobacterium tuberculosis causes tuberculosis, one of the world’s deadliest diseases. It is an acid-fast bacillus that is 2-4 um in length and 0.2-0.5 um in width. How many weeks, days,& hours would it take to see a M.tuberculosis colony on solid media? Its generation time is listed in the reading (above). Assume that bacterial colonies are not visible to the naked eye unless they contain ≥ 2 millioncells. This species forms aggregates of 5 cells on average. Show your...
