Q.11. From a skin biopsy a fibroblasts culture was taken .

Fibroblasts is cultured in exogenous sulfatides presence.

PROCEDURE Method 1. :- 1. from person having genetiv abnormalities skin fibtoblast cell is cultured

2.. it is done in liquid nitrogen through in vitero aging

3 . The cells are grown in fresh medium for freezing

4.. after 24 hrs tey are taken out with EDTA TRYPSIN. This is done because of suspended in growth medium .

5.. trypan blue dye test is done with 0.5 ml aliquot.

6.. centrifugation occur for five minutes at 900 rpm.

PROCEDURE Method. 2 :-

1.. at 37 degree centigrade in medium fibroblast are cultured in a humitified atmosphere of 5% co2 .

3.. with 0.25% trypsin or EDTA fibroblast are trypsinized sequentially .

4.. for cell growth and viability cell coubting method is used .

5... for storing of cell freezing medium is used that consists of 40% FCS

Result :- we examined primary edult scheme fibroblast.

2.. after 4 to 12 days outgrowth of cells are seen.

3.. at around 7 to 10 days appearances of fibroblasts are seen.
