2) Enzyme activity was measured to indidirectly measure the rate of substrate consumption.

3) True. As substrate concentration is an essential parameter to determine enzyme activity, once the enzyme is saturated with certain substrate concentration, further addition of substrate or increase in concentration of substrate does not alter the rate of the reaction.
