Answer #1

B. A low nutrient or inhibitory/selective medium like R2A,EMB or PIA are various types of bacterial culture medium. which are use as a selective medium so they are not ideal culture medium. So these medium will slows down the growth of the bacterial culture population. The lag phase will be too much long.

A. MacFarland is a standarized sol. used to compare the density of the bacterial population by using the absotbace value.

A haploid strain of yeast is treated with a mutagen called EMS. The different steps in the treatment and the analysis are the following: Step 1: Two tubes, labeled A and B contain 0.2 ml each of same yeast liquid culture. The concentration of yeast cells in the liquid culture was determined by a measure of the turbidity of the culture. This concentration is 4.4 x 109 cells/ml. Step 2: In each tube (A and B) 1.5 ml of phosphate...

You are given a mixed culture of two organisms in a small tube of TSB. These two organisms include a Gram-negative strain (your bacterial strain to identify) and a Gram-positive strain (a contaminant). Your first task is to isolate these organisms which will help you identify which Gram-negative strain you are working with. 1) You inoculate two streak plates on TSA media and MacConkey Agar using aseptic technique. For each medium, state whether the medium is selective, differential, or neither and...

This was followed by a subculturing step where each initial culture was used to inoculate a sterile nutrient-rich medium that contained NO antibiotic. 6. Transposon mutagenesis was performed to identify genes that are involved in the biosynthesis of theA amino acid alanine. A wildtype culture of E. coli was infected with a defective phage carrying Tn10 A and the culture was plated on LB-tet media to obtain tet-resistant colony forming units. (LB is a rich growth media for E. coli and contains all amino acids required for bacterial growth) Individual CFU were then transferred to a multiwell plate containing liquid LB-tet media and...
