When we add phenol chloroform to the isolation mixture during DNA extraction, than two layers are formed, upper aqueous layer and lower organic layer.

The lower organic layer contains all the biomolecules but the upper aqueous layer contains DNA which is solubilized into it.

The aqueous layer is than transferred to a new tube amd ethanol is added to precipitate out DNA.
