Answer #1

NB disc sensitivity test is performed inorder to identify Staphylococcus saprophyticus infection which is coogulase negative.

one of the main pathogen that causes UTI is S saprophyticus. Performing coogulase test can help us identify if the pathogen is coogulase positive Staphylococcus aureus or negative species. Among coogulase negative- the suspected bacteria can be either S saprophyticus or S epidermidis which is normal flora. so to differentiate them Novobiocin disc sensitivity test is performed. This disc is placed on blood agar which has inoculated pathogen. It is to check if the bacteria is sensitive or resistant to it. S saprophyticus is sensitive whereas others are resistant.

S epidermidis- resistant to NV and so shows no zone

S saprophyticus is sensitive and therefore shows zone around the disc.
