Cycle sequencing is an advanced method of nucleotide sequencing that utilizes a thermostable DNA polymerase and dideoxynucleotide triphosphates to generate sequences through chain termination with a temperature-cycling format. The cycle sequencing is certainly an improvement over conventional Sanger sequencing. The four ways are as following...

  • Cycle sequencing requires very less amount of template DNA comparing to that of Sanger sequencing
  • Adding fewer amount of template DNA ensures less impurities. So, the hazards related to pretreatment of template DNA is much lower
  • Multiple heat denaturation steps allow double stranded DNA templates to be sequenced efficiently without a separate denaturation step like in the Sanger sequencing. Thus the experiment takes less time to complete.
  • The thermostable DNA polymerase that is used Cycle sequencing is very much efficient in particular temperatures and synthesize new DNA strand more effectively compared to that of the normal DNA polymerase of Sanger sequencing. Cycle sequencing can sequence a lengthy DNA in less time comparing to the much more time taken with conventional Sanger sequencing.
